چکیده
Background aimsTo reduce the risk of graft-versus-host disease (GVHD) after allogeneic stem cell transplantation (alloSCT), T-cell depletion (TCD) grafts can be performed by addition alemtuzumab (ALT) “to bag” (in vitro) before transplantation. In this prospective study, authors analyzed effect in vitro incubation with 20 mg ALT on composition prior to graft infusion. Furthermore, assessed whether at moment infusion was predictive for reconstitution and development GVHD early TCD alloSCT.MethodsSixty granulocyte colony-stimulating factor-mobilized were obtained from ?9/10 HLA-matched related unrelated donors. The flow cytometry ALT. incidence severe monitored until 12 weeks transplantation.ResultsIn resulted an initial median efficiency receptor (TCR) ?/? T cells 96.7% (range, 63.5–99.8%), followed subsequent vivo. Graft volumes absolute leukocyte counts not TCR depletion. CD4pos depleted more efficiently than CD8pos cells, naive regulatory memory effector cells. This differential subsets line their reported CD52 expression. efficiencies numbers (naive) or post- alloSCT.ConclusionsThe bag is easy, fast generally applicable strategy prevent patients receiving alloSCT myeloablative non-myeloablative conditioning because efficient donor-derived lymphocytes

Immune Reconstitution after Allogeneic Hematopoietic Stem Cell Transplantation
The timely reconstitution and regain of function of a donor-derived immune system is of utmost importance for the recovery and long-term survival of patients after allogeneic hematopoietic stem cell transplantation (HSCT). Of note, new developments such as umbilical cord blood or haploidentical grafts were associated with prolonged immunodeficiency due to delayed immune reconstitution, raising ...
متن کاملMonitoring of Pathogen-Specific T-Cell Immune Reconstitution after Allogeneic Hematopoietic Stem Cell Transplantation
The clinical outcome after allogeneic hematopoietic stem cell transplantation (HSCT) has been significantly improved during the last decades with regard to the reduction in organ failure, infection, and severe acute graft-versus-host disease. However, severe complications due to infectious diseases are still one of the major causes of morbidity and mortality after allogeneic HSCT, in particular...

BACKGROUND AND OBJECTIVES Bone marrow (BM) and blood stem cell (BSC) allografts differ considerably with respect to their content of progenitor cells and progenitor cell subsets as well as mature lymphocytes. The aim of this prospective, randomized study was to determine whether these differences have an impact on early post-transplant immune recovery. DESIGN AND METHODS In a prospective rand...

متن کاملmiR-625-3p is upregulated in CD8+ T cells during early immune reconstitution after allogeneic stem cell transplantation
Alloreactive CD8+ T-cells mediate the curative graft-versus-leukaemia effect, the anti-viral immunity and graft-versus-host-disease (GvHD) after allogeneic stem cell transplantation (SCT). Thus, immune reconstitution with CD8+ T-cells is critical for the outcome of patients after allogeneic SCT. Certain miRNAs such as miR-146a or miR-155 play an important role in the regulation of post-transpla...
